Fiber Photometry Recording System
Fiber photometry has become the technique of choice for many systems neuroscience researchers due to its relative simplicity compared to other in vivo recording techniques, its excellent signal-to-noise ratio, and its versatility across a wide range of behavioral paradigms. It is most commonly used in combination with genetically encoded calcium indicators (GECIs, such as the GCaMP series). GECIs alter their fluorescence intensity depending on whether or not they are bound to calcium. Since the intracellular calcium concentration in neurons is very tightly regulated and voltage-gated calcium channels open when a neuron generates an action potential, transient increases in intracellular calcium concentration—which lead to a transient increase in GECI fluorescence—can serve as a good indirect indicator of neuronal activity.
There are various photometry system models available, with the R821 being the most comprehensive.
Key characteristics of model R821:
Tricolor excitation:
- 410 nm (reference signal, for noise reduction)
- 470 nm (green fluorescence – GCaMP, dLight…)
- 560 nm (red fluorescence – RCaMP, jrGECO1a…)
Multichannel acquisition:
- Up to 9 simultaneous channels (multiple animals or multiple brain regions)
High-precision detection:
- Two high-sensibility detectors
- Reduced interference due to independent sequential detection
Flexible acquisition modes:
- Continuous, interal, triggered, delayed, programmed
Real-time display:
- Visualization of ΔF/F during the acquisition
Control and integration:
- Digital interface (4 inputs / 4 outputs)
- Compatible with optogenetics: stimulation + recording with the same fiber
Behavioral camera included:
- High definition
- Synchronized with the recording of fluorescence
Computer and software included:
- Plug-and-play software including experiment control, data acquisition, and analysis
